Can I custom make hoses for an application using the barbed fittings and hose clamps that are available for AC systems? Will that work ok with R134a or was that something that only worked with the lower pressures of R12? Would such a setup be reliable or should I have custom crimp-together hoses made?

Bard fittings and hose clamps with barrier hose is not a suggested option.

barbs are not pretty, but they can work, if no other option. I have a car with them- been many years. No problems at all. Pain to remove, though, if it ever came to it.

For the low side, I wouldn't argue. For the high side, any other hoses that use barbed fittings and hose clamps used for items intended to hold 300+ psi of pressure? Not being confrontational, just curious.
Barbed (R12) type fittings should never be utilized with barrier hose. The bards are deeper and tend to cut into the barrier and the result is a leaking hose. Worm clamps were never a good clamp to use on an AC system...yes, I know many were used....but just a personal opinion. Way back in the retro fit days we conducted multiple test with barbed fittings with worm clamps and standard (for the day) ferrule type clamps. The barrier hoses were all damaged due to the depth/cut of the barbed fittings.
